Yes, you can apply after B.Tech although you can give this exam 4-5 times after B.Tech.

Clear GATE, state or institute level equivalent with a good score and you will become eligible for an M.Tech programme. Some companies accept GATE scores for their recruitment process.

Admission to the M.Tech courses at IITs,NITs,etc So very clearly, one can only sit for the GATE Exam after B.Tech as it is the entrance for the corresponding master course, I.e M.Tech.
