Can I sit for JEE Mains exam after completing Diploma?

Firstly, you can appear for JEE Mains exam on the basis of your Diploma also. Diploma is considered as equivalent to class 12th by IIT, and hence you can indeed get admission into IIT based on your Diploma provided of course you have fulfilled other associated criteria. https://www.jee-main.net/eligibility-criteria-2019/

The diploma students can get admission into IITs alone and not in NITs, IIITs or GFTIs. They can appear for JEE Mains and need to qualify it for getting eligible to write JEE Advanced.
to appear in JEE (Advanced) a diploma holder can apply in JEE Main but
his rank and score for JEE (Main) will not be declared as diploma holders cannot apply for admission in NITs and CFTIs through JEE (Main) channel.
