I am in 12th (commerce) and I want to open my own office for print media. Do I need B.Com degree for the same?

If you want to start your business, then it can be started after your 12th also but to successfully run a business, one should have the complete knowledge of the field. So, if you pursue graduation in Print media like B.Com or B.Sc that would be helpful as you will the basic understanding of the fie

Yes, you can go for direct course in journalism. You can do BA in Mass Media or Journalism.
