I am pursuing Diploma 2 second year can I take admission in B.Tech in ECE stream as distance course.

B.Tech through distance mode is not valid, so never be pursue. If you have passed the 12th than you can register for Section-A of AMIE as Technician member. Otherwise register after completion of Diploma course as Senior Technician Member. So the AMIE is the best for Diploma engineers who wants to u

Yes, after you complete your Diploma, definitely you can take lateral entry. What Mr Pradeep Kumar, has advised is absolutely valid. Just donot think of engineering via distance education.
