Yes, you can take admission in 2nd PUC after completing a Diploma course in Karnataka. The Department of Pre-University Education (DPUE) in Karnataka allows students to take admission in 2nd PUC through Lateral Entry Admission (LEA).

To be eligible for LEA, you must have completed a Diploma course in a relevant field from a recognised institution. You must also have secured at least 45% marks in the aggregate in your Diploma course.
