Is it necessary to do B.Tech to get into ISRO? Which are the ideal colleges/branches I should opt for?

It is not necessary to do B.Tech though it will raise your chances of getting into it. Through B.Tech, one need to have their stream of aeronautical and aerospace engineering. I won't say it is necessary to do a B.Tech but it would tremendously increase your chances to get into ISRO. Best option would be getting into IIST (Indian Institute of Space technology).
