    No, it's not necessary to have maths in intermediate in order to pursue your desired subject.

    No, it can't be necessary for you to study Maths. I want to suggest to you that, if you want to go in M.Sc Biotechnology, then go for B.Sc Biotech. So that, you can easily get admission in M.Sc program for biotech.
