NCERT (National Council of Educational Research and Training) textbooks from classes 11th and class 12th are the main source of questions in the Indian Institute of Science Education and Research Aptitude Test (IISER IIA). However, students can refer to other specialised books after preparing from National Council of Educational Research and Training textbooks. For more information, you may visit the Shiksha IISER exam page. (https://www.shiksha.com/college/iiser-Bhopal-indian-institute-of-science-education-research-57301). I hope this will help you.

Yes if you have read NCERT without skipping any line that's enough now you only need practice previous year questions and most important mocks with analysis. Give a mock test and analyse which portion you have not command and again go through it you can alse take the help of YouTube videos for any specific topic.
