Yes, Your bother will be eligible to get a seat through JEE Main if he clears his Class 12th supplementary exams. Clearing of Class 12 is a mandatory requirement. Validity of supplementary exams will depend upon clearing the subjects required.
For admission to NITs, IIITs, and GFTIs, he must have obtained at least 75% marks in Class 12 or must be within the top 20 percentile of his board (65% for SC/ST).
