Hello Aman!
No. JEE is an all India examination which would provide you an opportunity to study engineering in the prestigious Institutes like NITs and IITs.
But it is not compulsory.
You may also go for other universities like BITS-Pilani, VIT, etc. by clearing the respective exams or a better college with your board exam marks.

Both M.Tech and MBA are good career options after engineering graduation degree. M.Tech will make you a specialist in chosen field and you can make career as a technologist. You may seek admission in M.Tech if you are technically inclined. If you are not technically inclined then you may opt for MBA and become a generalist. I haven't attended IISER entrance exam. But I can let you know what my friends who wrote and got admitted to IISER have told me.
A first and most important thing you should have cleared DST-INSPIRE cutoff, which is different for different boards.
Exam is not going to very tough but may be bit tricky or challenging. So, focus on basics. First start solving some selected problems from NCERT textbook, which are bit tricky or those one which you did not now solve in your first or second attempt. Then go for previous year IIT JEE (now JEE Advanced) and AIEEE (now JEE main) papers. This much will do well.

Yes, you can appear in NEET there is no such conditions, you can check the website for more details but as you are already studying engineering and you are in 3rd year then why are you looking for any entrance exam like NEET.
You should concentrate on your engineering studies.

Distance Education B.Tech is not recognized as per the guidelines of AICTE and UGC. You can join as a lateral entry student in second year B.Tech CSE in any University or reputed College as a regular program.
