It entirely depends upon your choice as the three fields are entirely different from their own perspective.
If you want to carry on your study in the specialized field to get your Masters, then GATE is a good option.
If you want to join work and put to practice what you have learned, then IES would be perfect.
If you have managerial skills and want to get into management field then doing MBA would be great.

The dates for registration vary every year. But, generally the registration would commence by the end of November or from the beginning of December.
The last dates may extend for a month or more.
So, it is advisable to follow up every University's website periodically, so that you may not miss the dates.

As you are doing Diploma in mechanical, you have plenty of scope other than studying BE. If you are not interested in studying BE then it would better enough that you don't go for it.
As you have done Diploma, you can go for a job because, nowadays, there are more opportunities for the one who has completed Diploma in both private sectors as well as public sector.

Yes, scope is available, however, to get a job, you must be qualified and use the right network, securing a job immediately after your Master's degree may not be easy abroad, additional qualifications and/or work experience is recommended. With a microbiology degree, you can venture out into a wide variety of fields such as food technology, pharmacy, biotechnology, nanotechnology, clinical research etc. Research more by checking job portals abroad and look for related jobs, check university websites for research work, publications, etc., and you will have an idea about the scenario.
